The aim of this study was to develop a sensitive high-pressure liquid chromatographic (HPLC) method for the determination of the in vitro release of fluoride from a dental enamel matrix derivative (EMD). Release of fluoride from an EMD was followed by a separation of the fluoride compound on a microbore HPLC reverse-phase column using an ion-pairing mobile phase. Fluoride release was measured by loading two 10 mg EMD discs onto the surface of a 2.3 x 2.3 cm bovine enamel specimen. The fluoride released from the discs was measured with a pre-column stripping technique using ion-pairing HPLC. Enamel specimens were exposed to 37.50 microg ml(-1) fluoride solution for 15 min, removed from the solution and exposed to 25.00 microg ml(-1) fluoride solution for a second time. Following each cycle, the enamel was washed and dried for EMD sampling by the stripping method. The amount of fluoride released from the discs was determined by comparing the amount of fluoride remaining in the disc to the amount loaded onto the specimen. The HPLC method was linear from 5 to 50 microg ml(-1) and the limit of detection 3e33713323
